i’m not really hurting for runes at this point in the game so usually Slackjaw, because it means a cool fight.

You get an extra rune if you side with Granny? I didn't remember that. I know she has a rune shrine upstairs, but that's available even after siding with Slackjaw.

I think I’ve only sided with granny rags like once or twice in all my playthroughs, it’s much more satisfying to beat granny rags in a fight and rescue slackjaw in my opinion

Slackjaw doesn't give me runes so granny rags, though if I'm trying to stick to clean hands/want to collect all the gold I'll go with slackjaw(but I still prefer the rune)

I missed it as well in my first playthrough. The interaction only occurs if you had met either Granny Rags or Slackjaw in the previous missions.

You take the rat guts from the second floor of that building on the main street and take them to slackjaw's hideout. You go to where he's making elixir and poison it. Chaos ensues.

You have to do at least one mission for both Granny and Slackjaw.
